Immunoelectronmicroscopic analysis of surface antigens common to Mycobacterium bovis (BCG) and tumor cells.

چکیده
The presence of cell-surface antigens on line10 hepatocarcinoma cells was demonstrated by immunoelectronmicroscopy (IEM) with the use of xenogeneic and syngeneic tumor-specific antibody and syngeneic BCG-induced antibody. This confirmed that in addition to tumor-specific antigen(s) there are common antigens between line-10 hepatocarcinoma cells and BCG organisms. The specificity of this cross-reactivity was further supported by blocking experiments and absorption of the specific antibody to BCG organisms, with subsequent IEM of the BCG and loss of reactivity to tumor cells. Complement-dependent serum cytotoxicity was correlated with the IEM assay. Cross-reactivity of the BCG-specific antibody with the surface of cells of a human melanoma culture grown from an excised subcutaneous melanoma was also demonstrated by IEM.-J Natl Cancer Inst 53: 1313-1323, 1974.

Environmental strains of Mycobacterium avium interfere with immune responses associated with Mycobacterium bovis BCG vaccination.
Prior exposure of a vaccinee to certain species of environmental mycobacteria can prime the immune system against common mycobacterial antigens, which can in turn reduce the subsequent efficacy of live attenuated mycobacterial vaccines (such as Mycobacterium bovis BCG), in both human and livestock vaccination programs. In this study, two strains of Mycobacterium avium, both isolated from New Ze...
متن کاملImmunological evaluation of a component isolated from Mycobacterium bovis BCG with a monoclonal antibody to M. bovis BCG.
A component of Mycobacterium bovis BCG referred to as BCG-a was isolated through the combined use of monoclonal antibody directed to BCG and affinity chromatography. Analysis of BCG-a by sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed a single prominent band with a molecular weight of ca. 10,000. Structural characterization of BCG-a consisting of amino acid composition and am...
متن کاملCharacterization of fibronectin-binding antigens released by Mycobacterium tuberculosis and Mycobacterium bovis BCG.
Fibronectin (FN)-binding antigens are prominent components of short-term culture supernatants of Mycobacterium tuberculosis. In 3-day-old supernatants, a 30-kilodalton (kDa) protein was identified as the major FN-binding molecule. In 21-day-old supernatants, FN bound to a double protein band of 30 and 31 kDa, as well as to a group of antigens of larger molecular mass (57 to 60 kDa). FN-binding ...
متن کاملMycobacterium bovis Bacillus Calmette–Guérin Alters Melanoma Microenvironment Favoring Antitumor T Cell Responses and Improving M2 Macrophage Function
Intralesional Mycobacterium bovis bacillus Calmette-Guérin (BCG) has long been a relatively inexpensive therapy for inoperable cutaneous metastatic melanoma (CMM), although intralesional BCG skin mechanisms remain understudied. We analyzed intralesional BCG-treated CMM lesions combined with in vitro studies to further investigate BCG-altered pathways. Since macrophages play a pivotal role again...
متن کاملDevelopment of diagnostic reagents to differentiate between Mycobacterium bovis BCG vaccination and M. bovis infection in cattle.
In Great Britain a recent independent scientific review for the government has concluded that the development of a cattle vaccine against Mycobacterium bovis holds the best long-term prospect for tuberculosis control in British herds. A sine qua non for vaccination is the development of a complementary diagnostic test to differentiate between vaccinated animals and those infected with M. bovis ...
